Not sure that anyone will be able to assist with this but I thought I'd try.
My Grandfather's brother died at a relatively young age in 1930 in Dundee leaving a wife, three sons and a daughter. For some reason the children were all placed in care. The daughter was sent to a home in Aberdeen run by the RC Church, I believe the boys all went to somewhere in Edinburgh.
The three brothers are all deceased with no known descendants, their sister, now in advanced years has no knowledge of what happened to her brothers after the death of their father. There are no other family members that I am aware of who could help with this information.
Other than waiting for the 1931 census are there any other routes available to me to find out where the three boys went?

I am not sure what if any information you can obtain in relation to the children who were adopted.
Here are some dates instituted by Scotland in regard to the adoption act, Scotland was the first country in the UK to formalise adoptions.
Adoption of Children Act, 1926; Adoption of Children (Scotland) Act, 1930; the Children and Young Persons Act, 1933, and more.
It is possible the children's names were changed after adoption.

The children were never adopted, the deaths of the three brothers were registered under their names given at birth...

Because the events are relatively recent, I suspect you may have problems getting access to records, if records were made and if they have survived. I say this because at one point I was trying to find out some info about a childrens home in the early 1960s (elsewhere, not Edinburgh) and between records which were closed for 100years, and records which had not even been kept, I got nowhere. There may be records held in archives, but you may not be able to gain access to them.
My only suggestion is to contact Birthlink (in Edinburgh) who are an adoption charity but who also offer a service to those who have been in care, and they may just be able to give you some advice, even if they cannot help directly.
