Margaret Glacken born 1839 lanarkshire, father Hugh Glacken, mother Catherine ms McCartney.
I have documention from the Mitchell Libriary dated 1862 which states the family are getting poor relief, Margaret is married to James Todd with one daughter (not named). I have never been able to find any marriage details for Margaret and James, however, there is a a James Todd and a Margaret McLachlan married in 1860 Margarets Father is Hugh, mother Catherine ms McCartney. Is this just a coinsidence or is it possible the surname has been mistranscribed? Glacken and McLachlan soud very similar.

If all the other facts fit, it seem likely that the surname has been written down based on what someone thougth they heard. It happens quite often. And as you rightly say, if the name is said out loud, you can understand completely how the mistake may have been made.
(This is one area where us lot are often thought to be losing the plot, as our significant other walks past just as we are in the middle of conducting an experiment to better understand the situation, by saying these type of names out loud!
)
I may be remembering wrongly thre, but are these folk not also from Ireland? Chuck in an accent not from the local area and there is even more chance of the registrar not quite hearing it correctly.

I'd agree with Lesley including the talking out loud bit.
One way you might be able to double check is Margaret's address when she married. From the census records they did have a daughter - Margaret - about 1861/2 and went on to have lots more. Most of them are submissions on the IGI but Catherine born 1870 is a definite.

I have just found Agnes birth on ScotlandsPeople, parents James Todd and Margaret ms Glacken married 30th november 1859. James Todd and Margaret McLachlan were married 30th november 1860. I am pretty much convinced these are my folk.
Not sure about the address though Annette, this family moved around a lot. The only one I have ever found on the 1861 census is Margarets brother Christopher, he was an inmate at the parish poorhouse.

just to confuse this even more for me. I have just found James Todd's death. Parents are named as Andrew Todd , mother Margaret Steven, the marriage for James and Margaret McLachlan name his parents as Andrew and Janet Craig. Not sure any more. I think it's time for bed.

Who was the death informant? Is it his wife?
I have a similar case, also involving a couple from Ireland who came to Glasgow and married there in 1856 so they are about ages with your couple. The problem in my case I think, is that the parents of both parties remained in Ireland, and the wife who is stating the parents of her husband never met her in-laws...she is only going on what may have been mentioned a few times.
When he married, my man gave his parents as being Edward & Bridget, and some years after his death when applying for poor relief his wife gave his parents as being Peter and Margaret
...but she seems to have got the surnames correct in my case. (names were not given on his death - he died in the poorhouse, and the inforrmant was what appears to be another inmate.)
If your situation is similar, with the in-laws remaining in Ireland, I really do think it may be a symptom of just not knowing, because she never met them, and perhaps they were not spoken about much...? I would trust what your man said himself at the time of his marriage.
